Human Leukocyte Antigen

Histocompatibility agents (HLA, Human Leukocyte Antigen) are considered among the most polymorphic and clinically significant in immunogenetics. HLA class II (DQA1, DQB1, DRB1) genotyping is of vast importance in ensuring transplant compatibility, autoimmune diseases and reproductive disorders diagnosis. PCR testing offers high analytical sensitivity and specificity and ensures high speed of testing.

The HLA-DQA1 gene is characterized by a high degree of polymorphism, with currently more than 70 known alleles grouped into 8 main groups. Genotyping of DQA1 is performed at the allele group level (low resolution), enabling the selection of tissue-compatible donors for organ and tissue transplantation, the determination of genetic predisposition to autoimmune diseases, and the diagnostic evaluation of reproductive disorders.

The HLA-DQB1 gene is one of the most polymorphic within the HLA class II complex, comprising 12 allele groups. Its genotyping is also utilized in transplantology and immunogenetics, as its variability determines the immune response and the risk of developing autoimmune diseases, including celiac disease and type 1 diabetes mellitus.

The HLA-DRB1 gene exhibits a pronounced level of polymorphism, with over 3000 alleles divided into 13 groups. Genotyping of DRB1 is used to predict the development of various autoimmune diseases, including rheumatoid arthritis, multiple sclerosis, and systemic lupus erythematosus. In transplantology, this gene plays a crucial role in selecting tissue-compatible donors and predicting the risk of graft rejection.

Genotyping of HLA-DQA1, HLA-DQB1, and HLA-DRB1 is widely used in clinical practice.

Indications

  • Selection of tissue-compatible donors and recipients for organ and tissue transplantation
  • Screening of potential hematopoietic stem cell donors
  • Assessment of transplant rejection risk
  • Differential diagnosis and prognosis of autoimmune disease development
  • Diagnosis of reproductive disorders of unknown etiology

REAGENT KIT DESIGNED FOR HLA CLASS II DQA1 GENE TYPING BY REAL-TIME PCR

The HLA-DQA1 reagent kit is designed for the determination of human major histocompatibility complex specificities at the gene level by polymerase chain reaction in human biological material using detecting thermal cycler. The obtained results can be used for selecting a histocompatible donor for organ and tissue transplantation, and for the genetic prediction of immune-mediated diseases and reproductive disorders.

The reagent kit is designed for the simultaneous detection of 8 alleles and allele groups of the DQA1 gene (DQA10101, *0102, *0103, *0201, *0301, *0401, 0501, 0601) of the human major histocompatibility complex.

The kit can be used in clinical diagnostic laboratories of medical institutions and in research practice.

The real-time format is designed for PCR results detection during amplification using detecting thermal cyclers.
